Silence Therapeutics Hits the Catalyst Zone—Divesiran Data Could Send SLN Into Overdrive

The randomized, double-blind, placebo-controlled Phase 2 SANRECO study enrolled 48 phlebotomy-dependent patients, with the primary endpoint focused on maintaining hematocrit below 45% without phlebotomy between weeks 18 and 36.

The setup heading into the readout is notable because earlier data have already given investors something to watch. At the European Hematology Association 2026 Congress, Silence reported follow-up Phase 1 results showing that only five phlebotomies were required during active treatment among 21 patients, compared with 80 in the six months before treatment. Only four additional phlebotomies were reported during the 16-week post-treatment follow-up, while the company also reported improvements in PV-related symptoms and quality of life. No dose-limiting toxicities were observed in that Phase 1 analysis.

The Phase 2 data now have the potential to determine whether those earlier signals translate into controlled clinical evidence. Investors will likely focus heavily on hematocrit control, phlebotomy reduction, safety and the performance of the Q6W and Q12W dosing regimens. A convincing result could strengthen the case for divesiran as a differentiated treatment approach in PV and potentially elevate Silence's broader mRNAi GOLD platform. Conversely, as with any clinical-stage biotech, disappointing efficacy or safety results could materially change the investment thesis.
